چکیده

The transport systems for a-AIB and neutral, acid, and basic amino acids into lens were found to comprise a ~N a*-dependent and a Na-independent component. Na ions, required for 60 to 70 per cent of C-a-AlB transported into rabbit lens, were combined with a-AIB molecules in a 1:1 ratio while transported by a common "carrier" system. However, Na ions did not affect the diffusion constant nor the C-a-AIB efflux rates from the lens. The in vitro lens incubations showed accumulation by the lens and against the concentration gradient of >C-aAIB and other >C-amino acids. Throughout the incubation periods, Na ions transported with the aynino acid continued to be actively extruded from the lens. The Na+ deprivation effects closely resembled ouabain inhibitory effects on a-AIB transport, and were both exerted in the lens epithelium. These remits, and the independence of the lens a-AIB and K or Rb carriers, established the presence of a Na ions link between the IS!a*-dependent fraction of a-AIB and other amino acid transport systems and the ouabain-sensitive Na+-K "pump."

Discrimination of Single Transport Systems

Uptake of methionine, alpha-aminoisobutyric acid, and alpha-(methyl-amino)-isobutyric acid has been shown to occur by at least two transport systems, one sensitive and the other insensitive to the Na(+) concentration. For alpha-aminoisobutyric acid and its N-methyl derivative, the Na(+)-insensitive uptake is not concentrative and its rate increases almost linearly with concentration within the ...

Characterization of neutral amino acid transport in a marine pseudomonad.

The transport of neutral amino acids in marine pseudomonad B-16 (ATCC 19855) has been investigated. From patterns of competitive inhibition, mutant analysis, and kinetic data, two active transport systems with overlapping substrate specificities were distinguished and characterized. One system (DAG) served glycine, D-alanine, D-serine, and alpha-aminoisobutyric acid (AIB) and, to a lesser exten...

Induction of amino acid transport in primary cultures of adult rat liver parenchymal cells by insulin.

Amino acid transport was studied in primary cultures of parenchymal cells isolated from adult rat liver by a collagenase perfusion technique and maintained as a monolayer in a serum-free culture medium. Amino acid transport was assayed by measuring the uptake of the nonmetabolizable amino acid, alpha-aminoisobutyric acid. Rat liver parenchymal cells transported alpha-aminoisobutyric acid by an ...

Active amino acid transport in plasma membrane vesicles from Simian virus 40-transformed mouse fibroblasts. Characteristics of electrochemical Na+ gradient-stimulated uptake.

Selectively permeable membrane vesicles isolated from Simian virus 40-transformed mouse fibroblasts catalyzed Na+ gradient-coupled active transport of several neutral amino acids dissociated from intracellular metabolism. Na+-stimulated alanine transport activity accompanied plasma membrane material during centrifugation in discontinuous dextran 110 gradients. Carrier-mediated transport into th...

Intraocular transport of myoinositol. II. Accumulation in the rabbit lens in vitro.

The characteristics of myoinositol transport in the lens were studied by culturing them in a medium containing the tritium-labeled compound. The accumulation in the lens takes place against a concentration gradient and is inhibited by ouabain, iodoacetate, phlorhizin, and at reduced temperature (Q,o = 3). The energy for transport is derived from glycolysis. The transport system follows Michaeli...
